Effects of irisin combined with extracorporeal shock wave on fracture healing in ovariectomized rats

    摘要:

    目的] 探究鸢尾素联合体外冲击波 (extracorporeal shock waves, ESW) 治疗去势大鼠骨质疏松性骨折的作用与机理。[方法] 80 只 SD 大鼠随机分为 5 组,每组 16 只。分别为假手术组、模型组、模型+ESW 组、模型+鸢尾素组、模型+ ESW+鸢尾素组。除假手术组仅行手术切口外,其他各组先后行双侧卵巢切除和左侧股骨骨折内固定,术后分别给予相应处理,其中,ESW 治疗每周 1 次,鸢尾素组骨折部位局部注射。治疗后 8 周处死动物,行骨密度、破骨细胞指数、micro-CT、 Western blot 和 RT-qPCR 检测。[结果] 在骨密度和破骨细胞指数均由优至劣低依次为,假手术组>模型+ESW+鸢尾素>模型+ ESW 组,或模型+鸢尾素组>模型组 (P<0.05)。MicroCT 检测,包括 BS/BV、Tb.N、Tb.Th 和 Tb.Sp,由优至劣低依次为,假手术组 > 模型+ESW+鸢尾素 > 模型+ESW 组,或模型+鸢尾素组>模型组 (P<0.05)。Western blot 检测 BMP2、 VEGF、Nrf2F 的 OH-1 的蛋白相对表达量由高至低依次为,假手术组>模型+ESW+鸢尾素>模型+鸢尾素组,或模型+ESW 组>模型组 (P< 0.05)。 RT-qPCR 检测 Nrf2 和 OH-1 mRNA 相对表达量由高至低依次为,假手术组>模型+ESW+鸢尾素>模型+鸢尾素组,或模型+ESW 组>模型组(P<0.05)。[结论]鸢尾素联合 ESW 可激活 Nrf2/OH-1 通路并促进 VEGF 和 BMP2 蛋白的表达,促进骨质疏松大鼠的骨折愈合。

    Abstract:

    [Objective] To explore the effect on mechanism of irisin combined with extracorporeal shock wave (ESW) on fracture heal- ing in the ovariectomized rats. [Methods] A total of 80 SD rats were randomly divided into 5 groups with 16 rats in each group, including sham operation group, model group, model +ESW group, model + irisin group, model +ESW+ irisin group. Except the animals in sham oper- ation group that received surgical incision only, all rates received bilateral ovariectomy, and then left femoral fracture and internal fixation successively, subsequently, corresponding postoperative treatments were given respectively, including ESW treatment once a week, and lo- cal injection of irisin group in fracture site. At 4 weeks after treatment, the animals were sacrificed for bone mineral density, osteoclast in- dex, microCT, Western blot and RT-QPCR assays. [Results] Bone mineral density and osteoclast index were ranked superior-inferiorly in orders of sham operation group > model + ESW + irisin group > model + ESW group or model + irisin group > model group (P>0.05) . Mi- croCT detection items, including BS/BV, Tb.N, Tb.Th and Tb also ranked superior-inferiorly in sequence of model + ESW+ irisin group > model +ESW group or model + irisin group > model group (P<0.05) . In term of Western blot detection, the relative protein expression of BMP2, VEGF, Nrf2F and OH-1 arranged up-down in order of sham operation group > model + ESW+ irisin group > model +ESW group or model + irisin group > model group (P<0.05) . In term of RT-QPCR assay, the relative mRNA expression of Nrf2 and OH-1 from high to low were: sham operation group > model +ESW+ irisin group > model + irisin group or model +ESW group > model group (P<0.05) . [Conclusion] Irisin combined with ESW might activate Nrf2/OH-1 pathway to promote the expression of VEGF and BMP2 proteins, and pro- mote fracture healing in the ovariectomized rats
