Abstract:[Objective] To explore the potential intervention mechanism of Yougui Wan on the deformed lumbar intervertebral disc structure in rats. [Methods] A total of 30 male Sprague-Dawley rats were randomly divided into 3 groups, with 10 rats in each group. The animals in the normal control group were healthy rats without special treatment, those in degeneration group were established the intervertebral disc degeneration model by puncturing the annulus. Rats in medical group were gavage Yougui Wan for 2 weeks on the basis of the degeneration group. Serum IL-10,MIF and TNF-α levels were measured by ELISA method, the protein expression levels of CollagenⅡ, Notch1 in intervertebral disc tissues were detected by Western Blot method. [Results] Compared with normal control group, IL-10, MIF and TNF-α levels in degeneration group and medical group were increased, the expression levels of collagenⅡ protein were decreased, and the expression levels of Notch1 protein were increased, the difference among all groups was statistically significant (P<0.05). Compared with the degeneration group, the levels of IL-10 in medical group were increased, the levels of MIF, TNF-α were decreased, the protein expression levels of collagenⅡ were increased, and the protein expression levels of Notch1 were decreased, the difference among all groups was statistically significant (P<0.05). [Conclusion] Yougui Wan can inhibit the inflammatory response of IDD rats, reduce the degradation of extracellular matrix, and alleviate intervertebral disc degeneration. The mechanism may be related to the regulation of Notch signaling pathway.
