    摘要:

    [目的]比较膝骨性关节炎(knee osteoarthritis, KOA)磨损软骨组织与相对正常软骨组织白细胞介素-1(interleukin1, IL-1)、白细胞介素-6 (interleukin-6, IL-6) 和骨形态发生蛋白 2 (bone morphogenetic protein 2, BMP2) mRNA 相对表达的差异。[方法]选取本院行全膝关节置换术 KOA 12 例患者的磨损软骨样本(磨损组)和相对正常软骨样本(正常组)。提取软骨 mRNA,利用逆转录聚合酶链反应技术 (reverse transcription polymerase chain reaction, RT-PCR) 检测 IL-1、IL-6 和 BMP2 的 mRNA 的相对表达量,并进行两组间比较。[结果]磨损组的 IL-1 [(2.7±0.9) vs (0.2±0.1), P=0.013]、IL-6 [(7.2±2.0) vs (0.7±0.2), P= 0.004] 的 mRNA 相对表达量显著高于正常组,而磨损组 BMP2 的 mRNA 相对表达量显著低于正常组 [(0.4±0.2) vs (0.9±0.1), P= 0.015]。[结论]KOA 磨损软骨中 IL-1、IL-6 处于高表达状态,而 BMP2 却处于低表达状态,这表明 KOA 的软骨损害过程中软骨的分解过程强于合成过程。

    Abstract:

    [Objective] To compare mRNA relative expression of interleukin-1 (IL-1), interleukin-6 (IL-6) and bone morphogenetic protein 2 (BMP2) between the worn cartilage and relatively normal cartilage in knee osteoarthritis (KOA). [Methods] The worn cartilage samples (the worn group) and relatively normal cartilage samples (normal group) were harvested from 12 patients who underwent total knee arthroplasty for KOA in our hospital. After cartilage mRNA was extracted, the relative expression levels of mRNA of IL- 1, IL-6 and BMP2 were detected by reverse transcription polymerase chain reaction (RTPCR), and compared between the two groups. [Results] The worn group proved significantly higher relative expression of mRNA than the normal group in terms of IL-1 [(2.7±0.9) vs (0.2±0.1), P=0.013] and IL-6 [(7.2±2.0) vs (0.7±0.2), P=0.004], whereas the former was significantly lower than the latter regarding relative expression of mRNA of BMP2 [(0.4±0.2) vs (0.9±0.1), P=0.015]. [Conclusion] In the worn cartilage of KOA, the IL-1 and IL-6 are in a highly expressed state, while BMP2 is in a low-expressed state, which implies that the decomposition process of cartilage is stronger than the synthesis process during the cartilage damage process in KOA.
